Mutants lacking functional HYD2 homoeologs showed improved seedling growth, but comparable or increased susceptibility to salt stress in tillering plants, suggesting a developmentally restricted role of HYD2 in salt response. Salinity stress threatens global food security by reducing the yield of staple crops such as wheat (Triticum ssp.). Understanding how wheat responds to salinity stress is crucial for developing climate resilient varieties. In this study, we examined the interplay between carotenoid metabolism and the response to salt (NaCl) stress, a specific form of salinity stress, in tetraploid wheat plants with mutations in carotenoid β-hydroxylase 1 (HYD1) and HYD2. Our investigation encompassed both the vulnerable seedling stage and the more developed tillering stage of wheat plant growth. Mutant combinations lacking functional HYD2 homoeologs, including hyd-A2 hyd-B2, hyd-A1 hyd-A2 hyd-B2, hyd-B1 hyd-A2 hyd-B2, and hyd-A1 hyd-B1 hyd-A2 hyd-B2, had longer first true leaves and slightly enhanced root growth during germination under salt stress compared to the segregate wild-type (control) plants. Interestingly, these mutant seedlings also showed decreased levels of neoxanthin and violaxanthin (xanthophylls derived from β-carotene) and an increase in β-carotene in roots. However, tillering hyd mutant and segregate wild-type plants generally did not differ in their height, tiller count, and biomass production under acute or prolonged salt stress, except for decreases in these parameters observed in the hyd-A1 hyd-B1 hyd-A2 hyd-B2 mutant that indicate its heightened susceptibility to salt stress. Taken together, these findings suggest a significant, yet developmentally restricted role of HYD2 homoeologs in salt-stress response in tetraploid wheat. They also show that hyd-A2 hyd-B2 mutant plants, previously demonstrated for possessing enriched nutritional (β-carotene) content, maintain an unimpaired ability to withstand salt stress.

Wheat is an important contributor to global food security, and further improvements are required to feed a growing human population. Functional genetics and genomics tools can help us to understand the function of different genes and to engineer beneficial changes. In this study, we used a promoter capture assay to sequence 2-kb regions upstream of all high-confidence annotated genes from 1,513 mutagenized plants from the tetraploid wheat variety Kronos. We identified 4.3 million induced mutations with an accuracy of 99.8%, resulting in a mutation density of 41.9 mutations per kb. We also remapped Kronos exome capture reads to Chinese Spring RefSeq v1.1, identified 4.7 million mutations, and predicted their effects on annotated genes. Using these predictions, we identified 59% more nonsynonymous substitutions and 49% more truncation mutations than in the original study. To show the biological value of the promoter dataset, we selected two mutations within the promoter of the VRN-A1 vernalization gene. Both mutations, located within transcription factor binding sites, significantly altered VRN-A1 expression, and one reduced the number of spikelets per spike. These publicly available sequenced mutant datasets provide rapid and inexpensive access to induced variation in the promoters and coding regions of most wheat genes. These mutations can be used to understand and modulate gene expression and phenotypes for both basic and commercial applications, where limited governmental regulations can facilitate deployment. These mutant collections, together with gene editing, provide valuable tools to accelerate functional genetic studies in this economically important crop.

Many resistance genes deployed against pathogens in crops are intracellular nucleotide-binding (NB) leucine-rich repeat (LRR) receptors (NLRs). The ability to rationally engineer the specificity of NLRs will be crucial in the response to newly emerging crop diseases. Successful attempts to modify NLR recognition have been limited to untargeted approaches or depended on previously available structural information or knowledge of pathogen-effector targets. However, this information is not available for most NLR-effector pairs. Here, we demonstrate the precise prediction and subsequent transfer of residues involved in effector recognition between two closely related NLRs without their experimentally determined structure or detailed knowledge about their pathogen effector targets. By combining phylogenetics, allele diversity analysis, and structural modeling, we successfully predicted residues mediating interaction of Sr50 with its cognate effector AvrSr50 and transferred recognition specificity of Sr50 to the closely related NLR Sr33. We created synthetic versions of Sr33 that contain amino acids from Sr50, including Sr33syn, which gained the ability to recognize AvrSr50 with 12 amino-acid substitutions. Furthermore, we discovered that sites in the LRR domain needed to transfer recognition specificity to Sr33 also influence autoactivity in Sr50. Structural modeling suggests these residues interact with a part of the NB-ARC domain, which we named the NB-ARC latch, to possibly maintain the inactive state of the receptor. Water stress is a major obstacle to agricultural production, significantly impacting both yield and quality. During the 2017–18 crop year in Shahriar region near Tehran, Iran, known for its dry and cold climate, a study was conducted to examine the influence of different irrigation levels on the quantitative and qualitative characteristics of various wheat cultivars. The experiment involved three irrigation levels: normal irrigation (control), withholding irrigation at the flowering stage, and withholding irrigation at the seed-filling stage as primary factors, with 21 different cultivars as secondary factors. The analysis showed that irrigation, cultivar type, and their interactions had a significant effect on grain yield, proline, total chlorophyll, carbohydrate content in the first and second internodes during flowering and ripening, as well as on SOD, CAT, GPX, MDA, DT, and D-OH-dG levels at a one percent significance level. Withholding irrigation at the flowering stage had a more severe impact compared to the seed-filling stage. The Rakhshan and Sivand cultivars yielded the highest and lowest grain yields under normal irrigation conditions, while the drought-tolerant Ofogh cultivar showed lower yields when irrigation was withheld at the flowering stage. Water stress led to increased proline levels and higher levels of SOD, CAT, MDA, DT, and D-OH-dG in wheat cultivars. The Rakhshan and Sivand cultivars were identified as drought-tolerant in this region.

Abstract Fusarium head blight (FHB) is a significantly important disease in cereals primarily caused by Fusarium species. FHB control is largely executed through chemical strategies, which are costlier to sustainable wheat production, resulting in leaning towards sustainable sources such as resistance breeding and biological control methods for FHB. The present investigation was aimed at evaluating newly identified bacterial consortium (BCM) as biocontrol agents for FHB and understanding the morpho-physiological traits associated with the disease resistance of spring wheat. Preliminary evaluation through antagonistic plate assay and in vivo assessment indicated that BCM effectively inhibited Fusarium growth in spring wheat, reducing area under disease progress curve (AUDPC) and deoxynivalenol (DON), potentially causing type II and V resistance, and improving single spike yield (SSPY). Endurance to FHB infection with the application of BCM is associated with better sustenance of spike photosynthetic performance by improving the light energy harvesting and its utilization. Correlation and path-coefficient analysis indicated that maximum quantum yield (QY_max) is directly influencing the improvement of SSPY and reduction of grain DON accumulation, which is corroborated by principal component analysis. The chlorophyll fluorescence traits identified in the present investigation might be applied as a phenotyping tool for the large-scale identification of wheat sensitivity to FHB.

Allelo-chemical has been emerged as an important play to induce the abiotic stress tolerance. The experiment included three components: different levels of salinity stress (SS: control, 6 dS m-1, 12 dS m-1), seed priming with moringa leaf extract (MLE: 0.5%, 1.0%, 1.5%, 2.0%, 2.5%, 3.0%), and saltwater-tolerant and salinity-sensitive wheat cultivars (Faisalabad-2008, Galaxy-2013). Results showed that salinity lowered photosynthetic pigments, photosynthesis, transpiration, internal carbon, and stomatal conductance, while causing poor and delayed germination, inconsistent seedling growth, and increased hydrogen peroxide accumulation. However, hydro-priming and MLE priming enhanced emergence dynamics, growth, biochemical and enzymatic characteristics, and physiological aspects. The cultivar Faisalabad-2008 (wheat) performed well, but at high salinity levels, the hormetic impact of moringa leaf extract was more obvious, enhancing the germination and growth of cultivar Galaxy-2013, which was salinity-sensitive. Wheat cultivars' germination and seedling growth improved most when primed with 2% MLE (Faisalabad-2008) and 2.5% MLE (Galaxy-2013). This demonstrated that moringa possesses growth-promoting compounds that efficiently mitigate toxic impacts of salinity.

Abstract Background Fructans are water-soluble carbohydrates that accumulate in wheat and are thought to contribute to a pool of stored carbon reserves used in grain filling and tolerance to abiotic stress. Results In this study, transgenic wheat plants were engineered to overexpress a fusion of two fructan biosynthesis pathway genes, wheat sucrose: sucrose 1-fructosyltransferase (Ta1SST) and wheat sucrose: fructan 6-fructosyltransferase (Ta6SFT), regulated by a wheat ribulose-1,5-bisphosphate carboxylase/oxygenase small subunit (TaRbcS) gene promoter. We have shown that T4 generation transgene-homozygous single-copy events accumulated more fructan polymers in leaf, stem and grain when compared in the same tissues from transgene null lines. Under water-deficit (WD) conditions, transgenic wheat plants showed an increased accumulation of fructan polymers with a high degree of polymerisation (DP) when compared to non-transgenic plants. In wheat grain of a transgenic event, increased deposition of particular fructan polymers such as, DP4 was observed. Conclusions This study demonstrated that the tissue-regulated expression of a gene fusion between Ta1SST and Ta6SFT resulted in modified fructan accumulation in transgenic wheat plants and was influenced by water-deficit stress conditions.

A two-year field experiment was conducted to measure the effects of densification methods on photosynthesis and yield of densely planted wheat. Inter-plant and inter-row distances were used to define rate-fixed pattern (RR) and row-fixed pattern (RS) density treatments. Meanwhile, four nitrogen (N) rates (0, 144, 192, and 240 kg N ha−1, termed N0, N144, N192, and N240) were applied with three densities (225, 292.5, and 360 × 104 plants ha−1, termed D225, D292.5, and D360). The wheat canopy was clipped into three equal vertical layers (top, middle, and bottom layers), and their chlorophyll density (ChD) and photosynthetically active radiation interception (FIPAR) were measured. Results showed that the response of ChD and FIPAR to N rate, density, and pattern varied with different layers. N rate, density, and pattern had significant interaction effects on ChD. The maximum values of whole-canopy ChD in the two seasons appeared in N240 combined with D292.5 and D360 under RR, respectively. Across two growing seasons, FIPAR values of RR were higher than those of RS by 29.37% for the top layer and 5.68% for the middle layer, while lower than those of RS by 20.62% for the bottom layer on average. With a low N supply (N0), grain yield was not significantly affected by density for both patterns. At N240, increasing density significantly increased yield under RR, but D360 of RS significantly decreased yield by 3.72% and 9.00% versus D225 in two seasons, respectively. With an appropriate and sufficient N application, RR increased the yield of densely planted wheat more than RS. Additionally, the maximum yield in two seasons appeared in the combination of D360 with N144 or N192 rather than of D225 with N240 under both patterns, suggesting that dense planting combined with an appropriate N-reduction application is feasible to increase photosynthesis capacity and yield.

Abstract Background Class III peroxidases (PODs) perform crucial functions in various developmental processes and responses to biotic and abiotic stresses. However, their roles in wheat seed dormancy (SD) and germination remain elusive. Results Here, we identified a wheat class III POD gene, named TaPer12-3A, based on transcriptome data and expression analysis. TaPer12-3A showed decreasing and increasing expression trends with SD acquisition and release, respectively. It was highly expressed in wheat seeds and localized in the endoplasmic reticulum and cytoplasm. Germination tests were performed using the transgenic Arabidopsis and rice lines as well as wheat mutant mutagenized with ethyl methane sulfonate (EMS) in Jing 411 (J411) background. These results indicated that TaPer12-3A negatively regulated SD and positively mediated germination. Further studies showed that TaPer12-3A maintained H2O2 homeostasis by scavenging excess H2O2 and participated in the biosynthesis and catabolism pathways of gibberellic acid and abscisic acid to regulate SD and germination. Conclusion These findings not only provide new insights for future functional analysis of TaPer12-3A in regulating wheat SD and germination but also provide a target gene for breeding wheat varieties with high pre-harvest sprouting resistance by gene editing technology.

Abstract Stripe rust of wheat, caused by Puccinia striiformis f. sp. tritici (Pst), is one of the most important diseases of wheat worldwide. Identification of new and elite Pst-resistance loci or genes has the potential to enhance overall resistance to this pathogen. Here, we conducted an integrated genome-wide association study (GWAS) and transcriptomic analysis to screen for loci associated with resistance to stripe rust in 335 accessions from Yunnan, including 311 landraces and 24 cultivars. Based on the environmental phenotype, we identified 113 protein kinases significantly associated with Pst resistance using mixed linear model (MLM) and generalized linear model (GLM) models. Transcriptomic analysis revealed that 52 of 113 protein kinases identified by GWAS were up and down regulated in response to Pst infection. Among these genes, a total of 15 receptor kinase genes were identified associated with Pst resistance. 11 candidate genes were newly discovered in Yunnan wheat germplasm. Our results revealed that resistance alleles to stripe rust were accumulated in Yunnan wheat germplasm, implying direct or indirect selection for improving stripe rust resistance in elite wheat breeding programs.

Abstract Background B-box (BBX) proteins are a type of zinc finger proteins containing one or two B-box domains. They play important roles in development and diverse stress responses of plants, yet their roles in wheat remain unclear. Results In this study, 96 BBX genes were identified in the wheat genome and classified into five subfamilies. Subcellular localization prediction results showed that 68 TaBBXs were localized in the nucleus. Protein interaction prediction analysis indicated that interaction was one way that these proteins exerted their functions. Promoter analysis indicated that TaBBXs may play important roles in light signal, hormone, and stress responses. qRT-PCR analysis revealed that 14 TaBBXs were highly expressed in seeds compared with other tissues. These were probably involved in seed dormancy and germination, and their expression patterns were investigated during dormancy acquisition and release in the seeds of wheat varieties Jing 411 and Hongmangchun 21, showing significant differences in seed dormancy and germination phenotypes. Subcellular localization analysis confirmed that the three candidates TaBBX2-2 A, TaBBX4-2 A, and TaBBX11-2D were nuclear proteins. Transcriptional self-activation experiments further demonstrated that TaBBX4-2A was transcriptionally active, but TaBBX2-2A and TaBBX11-2D were not. Protein interaction analysis revealed that TaBBX2-2A, TaBBX4-2A, and TaBBX11-2D had no interaction with each other, while TaBBX2-2A and TaBBX11-2D interacted with each other, indicating that TaBBX4-2A may regulate seed dormancy and germination by transcriptional regulation, and TaBBX2-2A and TaBBX11-2D may regulate seed dormancy and germination by forming a homologous complex. Conclusions In this study, the wheat BBX gene family was identified and characterized at the genomic level by bioinformatics analysis. These observations provide a theoretical basis for future studies on the functions of BBXs in wheat and other species.

Abstract Background Ubiquitination is an important regulatory step of selective protein degradation in the plant UPS (ubiquitin–proteasome system), which is involved in various biological processes in eukaryotes. Ubiquitin-conjugating enzymes play an intermediate role in the process of protein ubiquitination reactions and thus play an essential role in regulating plant growth and response to adverse environmental conditions. However, a genome-wide analysis of the UBC gene family in wheat (Triticum aestivum L.) has not yet been performed. Results In this study, the number, physiochemical properties, gene structure, collinearity, and phylogenetic relationships of TaUBC family members in wheat were analyzed using bioinformatics methods. The expression pattern of TaUBC genes in different tissues/organs and developmental periods, as well as the transcript levels under abiotic stress treatment, were analyzed using RNA-Seq data and qRT-PCR. Meanwhile, favorable haplotypes of TaUBC25 were investigated based on wheat resequencing data of 681 wheat cultivars from the Wheat Union Database. The analyses identified a total of 93 TaUBC family members containing a UBC domain in wheat genome. These genes were unevenly distributed across 21 chromosomes, and numerous duplication events were observed between gene members. Based on phylogenetic analysis, the TaUBC family was divided into 13 E2 groups and a separate UEV group. We investigated the expression of TaUBC family genes under different tissue/organ and stress conditions by quantitative real-time PCR (qRT-PCR) analysis. The results showed that some TaUBC genes were specifically expressed in certain tissues/organs and that most TaUBC genes responded to NaCl, PEG6000, and ABA treatment with different levels of expression. In addition, we performed association analysis for the two haplotypes based on key agronomic traits such as thousand-kernel weight (TKW), kernel length (KL), kernel weight (KW), and kernel thickness (KT), examining 122 wheat accessions at three environmental sites. The results showed that TaUBC25-Hap II had significantly higher TKW, KL, KW, and KT than TaUBC25-Hap I. The distribution analysis of haplotypes showed that TaUBC25-Hap II was preferred in the natural population of wheat. Conclusion Our results identified 93 members of the TaUBC family in wheat, and several genes involved in grain development and abiotic stress response. Based on the SNPs detected in the TaUBC sequence, two haplotypes, TaUBC25-Hap I and TaUBC25-Hap II, were identified among wheat cultivars, and their potential value for wheat breeding was validated by association analysis. The above results provide a theoretical basis for elucidating the evolutionary relationships of the TaUBC gene family and lay the foundation for studying the functions of family members in the future.

Abstract Background Accuracy in determining food authenticity, possible contamination, content analysis, and even geographical origin is of considerable scientific and economic value. The aim of this study is to facilitate quantitative evaluation of protein content in the seeds of cereals (Triticum turgidum var. durum and Tritordeum genotypes) and ripening pomegranate fruits (Wonderful cultivar). Methods Two species of wheat were evaluated in this study: durum wheat, Triticum turgidum var. durum, and Tritordeum (durum wheat × wild barley) together with pomegranate fruits of the variety Wonderful. Two different portable Near InfraRed (NIR) spectrometers have been used: a prototype developed in the PhasmaFood project and the commercial SCiO™ molecular sensor. Results Considering the specific samples, the obtained results of the classification models indicate a validation mean absolute error of 0.8% (percentage of total protein content in dry matter) for two species of wheat using Convolutional Neural Network following normalization procedures and 0.32% using Partial Least Square (PLS) analysis applied to Tritordeum samples; visible reflectance spectra have been used to discriminate the two cereal species. A Root Mean Square Error (RMSE) of 1.25 was obtained for the determination of total soluble solids (TSS) over a 2-year period for pomegranate fresh fruits of Wonderful cultivar, which is commonly harvested with TSS values of 16–17. Conclusions The application of portable sensors using NIR spectroscopy can be a valid and rapid alternative to the use of destructive laboratory techniques for the assessment of protein content in intact wheat seeds and ripeness grade (TSS) in intact pomegranates.

Abstract Background Soil salinization is one of the vital factors threatening the world’s food security. To reveal the biological mechanism of response to salt stress in wheat, this study was conducted to resolve the transcription level difference to salt stress between CM6005 (salt-tolerant) and KN9204 (salt-sensitive) at the germination and seedling stage. Results To investigate the molecular mechanism underlying salt tolerance in wheat, we conducted comprehensive transcriptome analyses at the seedling and germination stages. Two wheat cultivars, CM6005 (salt-tolerant) and KN9204 (salt-sensitive) were subjected to salt treatment, resulting in a total of 24 transcriptomes. Through expression-network analysis, we identified 17 modules, 16 and 13 of which highly correlate with salt tolerance-related phenotypes in the germination and seedling stages, respectively. Moreover, we identified candidate Hub genes associated with specific modules and explored their regulatory relationships using co-expression data. Enrichment analysis revealed specific enrichment of gibberellin-related terms and pathways in CM6005, highlighting the potential importance of gibberellin regulation in enhancing salt tolerance. In contrast, KN9204 exhibited specific enrichment in glutathione-related terms and activities, suggesting the involvement of glutathione-mediated antioxidant mechanisms in conferring resistance to salt stress. Additionally, glucose transport was found to be a fundamental mechanism for salt tolerance during wheat seedling and germination stages, indicating its potential universality in wheat. Wheat plants improve their resilience and productivity by utilizing adaptive mechanisms like adjusting osmotic balance, bolstering antioxidant defenses, accumulating compatible solutes, altering root morphology, and regulating hormones, enabling them to better withstand extended periods of salt stress. Conclusion Through utilizing transcriptome-level analysis employing WGCNA, we have revealed a potential regulatory mechanism that governs the response to salt stress and recovery in wheat cultivars. Furthermore, we have identified key candidate central genes that play a crucial role in this mechanism. These central genes are likely to be vital components within the gene expression network associated with salt tolerance. The findings of this study strongly support the molecular breeding of salt-tolerant wheat, particularly by utilizing the genetic advancements based on CM6005 and KN9204.

Abstract Background Wheat is one of the world’s most important cereal crops. However, the fungal pathogen Zymoseptoria tritici can cause disease epidemics, leading to reduced yields. With climate change and development of new agricultural areas with suitable environments, Z. tritici may advance into geographical areas previously unaffected by this pathogen. It is currently unknown how Egyptian wheat will perform in the face of this incoming threat. This project aimed to assess the resistance of Egyptian wheat germplasm to Z. tritici, to identify cultivars with high levels of resistance and characterise the mechanism(s) of resistance present in these cultivars. Results Eighteen Egyptian wheat cultivars were screened against two Z. tritici model isolates and exhibited a wide spectrum of responses. This ranged from resistance to complete susceptibility to one or both isolates tested. The most highly resistant cultivars from the initial screen were then tested under two environmental conditions against modern UK field isolates. Disease levels under UK-like conditions were higher, however, symptom development on the cultivar Gemmeiza-12 was noticeably slower than on other Egyptian wheats. The robustness of the resistance shown by Gemmeiza-12 was confirmed in experiments mimicking Egyptian environmental conditions, where degree of Z. tritici infection was lower. The Kompetitive allele-specific PCR (KASP) diagnostic assay suggested the presence of an Stb6 resistant allele in several Egyptian wheats including Gemmeiza-12. Infection assays using the IPO323 WT and IPO323ΔAvrStb6 mutant confirmed the presence of Stb6 in several Egyptian cultivars including Gemmeiza-12. Confocal fluorescence microscopy demonstrated that growth of the IPO323 strain is blocked at the point of stomatal penetration on Gemmeiza-12, consistent with previous reports of Stb gene mediated resistance. In addition to this R-gene mediated resistance, IPO323 spores showed lower adherence to leaves of Gemmeiza-12 compared to UK wheat varieties, suggesting other aspects of leaf physiology may also contribute to the resistance phenotype of this cultivar. Conclusion These results indicate that Gemmeiza-12 will be useful in future breeding programs where improved resistance to Z. tritici is a priority.

Abstract. The effectiveness of mineral fertilization on wheat productivity was determined in a field experiment on Alluvial-meadow soil (Fluvisol, FAO 2015) at the experimental field of Tsalapitsa village, Plovdiv region, during 2019-2020. Four increasing rates of nitrogen fertilization along with a constant background of phosphorus fertilization were tested in the following variants: N40P100; N80P100; N120P100; N160P100, and N0P0 (control) – without any fertilization. The influence of these variants on yield and the content of basic macro- and microelements in wheat’s dry biomass were assessed. Plant samples were collected at the full maturity phase of wheat, with each variant sampled in triplicate, to determine the concentrations of N, P, K, Ca, Mg, Cu, Zn, Mn, and Fe. Correlation analysis and analysis of variance (ANOVA) were employed to identify dependencies between observed variables. Significant differences between the variants for N% content in grain, chaff and straw of wheat were established. The highest nitrogen values were observed in the grain of the N160P100 and N120P100 variants, at 2.10% and 2.02%, respectively. The applied fertilization had a significant positive effect on the accumulation of potassium, calcium, copper, manganese and iron in wheat biomass. This finding was supported by the correlation coefficients between fertilizer rates and N (R = 0.948), Ca (R = 0.854), Cu (R = 0.681), Mn (R = 0.735), and Fe (R = 0.769) in the wheat grain. Significant yield differences were observed among the variants, with the highest yield recorded for N120P100 (7123.0 kg/ha), more than tripling that of the control variant. The uptake of nitrogen, phosphorus and potassium by the total biomass of wheat increased depending on the fertilization rates and it was the highest in the N120P100 variant.

An Elite Spring Wheat Yield Trial (EWYT) was conducted with 50 diverse genotypes of Wheat (Triticum aestivum L.) and analyzed by multivariate analysis during 2018-19 rabi season. Correlation analysis revealed positive association of plant height, grains per spike, spike length, test weight and biomass with grain yield. Path analysis revealed the highest direct effect on grain yield/metre by biomass/metre (1.508) which was also positively associated with grain yield/metre (0.833). PCA analysis showed 11 principal components out of which five principal components (PC1 to PC5) exhibited eigen value greater than one. The correlation between AUDPC and physiological characters indicated that chlorophyll loss was associated with high spot blotch disease incidence at later stages of crop growth.

Crops are constantly exposed to pathogenic microbes. Rust fungi are examples of these harmful microorganisms, which have a major economic impact on wheat production. To protect themselves from pathogens like rust fungi, plants employ a multilayered immune system that includes immunoreceptors encoded by resistance genes. Significant efforts have led to the isolation of numerous resistance genes against rust fungi in cereals, especially in wheat. However, the evolution of virulence of rust fungi hinders the durability of resistance genes as a strategy for crop protection. Rust fungi, like other biotrophic pathogens, secrete an arsenal of effectors to facilitate infection, and these are the molecules that plant immunoreceptors target for pathogen recognition and mounting defense responses. When recognized, these effector proteins are referred to as avirulence (Avr) effectors. Despite the many predicted effectors in wheat rust fungi, only five Avr genes have been identified, all from wheat stem rust. Knowledge of the Avr genes and their variation in the fungal population will inform deployment of the most appropriate wheat disease-resistance genes for breeding and farming. The review provides an overview of methodologies as well as the validation techniques that have been used to characterize Avr effectors from wheat stem rust. Plant growth regulators (PGRs) play an important role in increasing crop yield, and quality, and enhancing crop stress resistance in agricultural production, especially for important crops. PGRs can affect the transport and distribution of assimilates by changing the content and distribution of endogenous hormones in plants. Numerous empirical research results have proven that PGRs have an important impact on the growth, development, and yield composition of wheat. Taking wheat plants as an example, this study reviews the application of PGRs in wheat production and explores their impact on wheat growth and yield. Furthermore, residues and microbial degradation of PGRs are summarized in detail. Finally, future research directions on PGR application in wheat production are proposed. This summary is of great significance for understanding the role of PGRs in wheat production.

Based on varieties, area and distribution of wheat in China, 1 377 newly harvested wheat samples from 12 provinces were collected in 2022. The physio-chemical quality as well as end-use quality evolutions were carried out, and the current quality status of wheat were analyzed. The results showed that the average crude protein content and gluten content of wheat were 13.1% and 29.6%, respectively, and the mean gluten index was 69. Among the rheological parameters of dough, the water absorption of flour was 63.2 mL, and the average stability time was 6.7 min. The mean tensile curve area, elongation and maximum tensile resistance were 75 cm2, 140 mm and 398 EU, respectively. All samples were evaluated for the processing quality of noodles and steamed bread, and samples with strong gluten wheat varieties and wet gluten content ≤25% were evaluated for the processing quality of bread and cake, respectively. The average scores of noodles, steamed bread, bread and cake were 79, 75, 73 and 69 points, respectively. Only 1.7% of the samples met the requirements of “High quality wheat-Strong gluten wheat” (GB/T 17892—1999), while 1.3% of the samples met the requirements of “High quality wheat-Weak gluten wheat” (GB/T 17893—1999). In addition, 44.9%, 31.4% and 2.8% of the samples met the requirements of quality classification, high-quality wheat and high-quality strong gluten wheat in “Quality classification of wheat varieties” (GB/T 17320—2013), respectively, and 56.0% and 3.0% of the samples met the requirements of quality classification and strong gluten hard wheat in “The Grain & Oil Products of China-Wheat” (LS/T 3109—2017), respectively. The results showed that the processing quality of wheat in China was general, and the food quality of steamed food was higher than that of baked food. A few varieties with excellent food processing quality bred were also planted.

Abstract In the modern era, intensive agricultural practices such as agrochemicals are applied in excessive amounts to enhance agricultural production. However, imbalanced adoption of these chemicals has arisen in the dwindling of agriculture factor productivity and soil quality. To maintain soil fertility and production, these chemical fertilizers must be supplemented with organic inputs. Keeping this in the backdrop, a research trail was established during 2018–19 and 2019–20 years at Research Farm of Agriculture University, Kota, India. The treatment setup was comprised of 5 treatment modules viz., conservation tillage + organic management (CAOM), conservation tillage + chemical management (CACM), conventional tillage + chemical management (CTCM), conventional tillage + organic management (CTOM) and the package of practices (PoPs) with four replications. Results indicated that the highest organic carbon (0.68%), bacterial (29.11 × 107 cfu g−1), fungal (4.77 × 104 cfu g−1), actinomycetes populations (5.67 × 104 cfu g−1), acid phosphatase (44.1 µg g−1 h−1), urease (45.3 µg g−1 h−1) and dehydrogenase (23.3 µg triphenylformazan [TPF] g−1 h−1) activity in soil were found in the treatment of conservation organic system during both the years of study at each soil depth. In contrast to other parameters, the highest system productivity was observed with conservation chemical crop management approaches, with a soybean equivalent yield of 4615 kg ha−1 in a soybean–wheat system of production. Furthermore, the soil quality index (SQI) significantly varied from the lowest score (0.30) at 45–60 cm layer of soil in the package of practices to the highest score (0.92) at 0–15 cm layer of soil with regards to the conservation organic which shows, 206.67 percent enhancement through the soil profile of various crop management practices. The SQI variation from 0–15 to 45–60 cm soil depth was 130.0, 81.08, 60.0, 175.0 and 83.33 percent, respectively, for CAOM, CACM, CTCM, CTOM and PoPs. Amongst, different systems, the highest mean performance was noticed under the conservation organic systems for physical and biological properties. Hence, in line with the salient outcome, we may propose that the conservation chemical system needs to be followed to improve crop productivity, whereas, conservation organic seems a good option for soil health with long-term viability.

Abstract To ascertain the ideal dosage of ZnO NPs (Zinc Oxide Nanoparticles), we conducted an investigation on the priming effects of varying concentrations of ZnO NPs on germination and physio-biochemical parameters of wheat. In this study, ZnO NPs were synthesized and characterized for their physico-chemical properties followed by confirmation of the formation of ZnO NPs. Throughout this study, wheat seeds were subjected to ZnO NPs at various concentrations of 5, 50, 100, 250, and 500 ppm for a period of 4 h via continuous aeration. The primed seeds were sowed in plastic bags, allowed to grow for 21 days, following which comprehensive evaluations of physio-biochemical attributes were conducted. At 250 ppm, an impressive 100% of seeds successfully germinated compared to the control group. The examined physiological factors such as shoot length, root length, and fresh as well as dry weights of leaf and root tissues all exhibited notable increases with the ascending concentrations of ZnO NP up to 250 ppm. However, beyond this threshold, at 500 ppm, these parameters experienced a decline. Inductively coupled plasma atomic absorption spectrophotometer (ICP-AAS) measurements validated the progressive increase in Zinc content in the nanoprimed seedlings, further affirming the dose-dependent trend. Zinc oxide nanoparticles notably improved key biochemical features, including elevated levels of total chlorophyll, malondialdehyde (MDA), total protein, and the accumulation of osmolytes such as proline and glycine-betaine. Additionally, the presence of ZnO NPs led to increased activity of antioxidant enzymes like superoxide dismutase (SOD) and catalase in a dose-dependent mananer. Collectively, the amassed data underscores the efficacy of the 250 ppm ZnO NPs treatment, which emerged as superior in comparison to both the control group and other administered treatments. These findings underscore the potential of ZnO NPs at a concentration of 250 ppm as a valuable seed nanopriming agent, effectively enhanced germination and robust early-stage growth in young plants.

Abstract Background RNA-seq is a fundamental technique in genomics, yet reference bias, where transcripts derived from non-reference alleles are quantified less accurately, can undermine the accuracy of RNA-seq quantification and thus the conclusions made downstream. Reference bias in RNA-seq analysis has yet to be explored in complex polyploid genomes despite evidence that they are often a complex mosaic of wild relative introgressions, which introduce blocks of highly divergent genes. Results Here we use hexaploid wheat as a model complex polyploid, using both simulated and experimental data to show that RNA-seq alignment in wheat suffers from widespread reference bias which is largely driven by divergent introgressed genes. This leads to underestimation of gene expression and incorrect assessment of homoeologue expression balance. By incorporating gene models from ten wheat genome assemblies into a pantranscriptome reference, we present a novel method to reduce reference bias, which can be readily scaled to capture more variation as new genome and transcriptome data becomes available. Conclusions This study shows that the presence of introgressions can lead to reference bias in wheat RNA-seq analysis. Caution should be exercised by researchers using non-sample reference genomes for RNA-seq alignment and novel methods, such as the one presented here, should be considered.

Abstract Background The severity and frequency of drought are expected to increase substantially in the coming century and dramatically reduce crop yields. Manipulation of rhizosphere microbiomes is an emerging strategy for mitigating drought stress in agroecosystems. However, little is known about the mechanisms underlying how drought-resistant plant recruitment of specific rhizosphere fungi enhances drought adaptation of drought-sensitive wheats. Here, we investigated microbial community assembly features and functional profiles of rhizosphere microbiomes related to drought-resistant and drought-sensitive wheats by amplicon and shotgun metagenome sequencing techniques. We then established evident linkages between root morphology traits and putative keystone taxa based on microbial inoculation experiments. Furthermore, root RNA sequencing and RT-qPCR were employed to explore the mechanisms how rhizosphere microbes modify plant response traits to drought stresses. Results Our results indicated that host plant signature, plant niche compartment, and planting site jointly contribute to the variation of soil microbiome assembly and functional adaptation, with a relatively greater effect of host plant signature observed for the rhizosphere fungi community. Importantly, drought-resistant wheat (Yunhan 618) possessed more diverse bacterial and fungal taxa than that of the drought-sensitive wheat (Chinese Spring), particularly for specific fungal species. In terms of microbial interkingdom association networks, the drought-resistant variety possessed more complex microbial networks. Metagenomics analyses further suggested that the enriched rhizosphere microbiomes belonging to the drought-resistant cultivar had a higher investment in energy metabolism, particularly in carbon cycling, that shaped their distinctive drought tolerance via the mediation of drought-induced feedback functional pathways. Furthermore, we observed that host plant signature drives the differentiation in the ecological role of the cultivable fungal species Mortierella alpine (M. alpina) and Epicoccum nigrum (E. nigrum). The successful colonization of M. alpina on the root surface enhanced the resistance of wheats in response to drought stresses via activation of drought-responsive genes (e.g., CIPK9 and PP2C30). Notably, we found that lateral roots and root hairs were significantly suppressed by co-colonization of a drought-enriched fungus (M. alpina) and a drought-depleted fungus (E. nigrum). Conclusions Collectively, our findings revealed host genotypes profoundly influence rhizosphere microbiome assembly and functional adaptation, as well as it provides evidence that drought-resistant plant recruitment of specific rhizosphere fungi enhances drought tolerance of drought-sensitive wheats. These findings significantly underpin our understanding of the complex feedbacks between plants and microbes during drought, and lay a foundation for steering “beneficial keystone biome” to develop more resilient and productive crops under climate change. The most important part of the plant antioxidant system is the ascorbate-glutathione cycle (AGC), the activity of which is observed upon exposure to a range of stressors, including lack of O2, and oxidative stress occurring immediately after the restoration of oxygen access, hereafter termed reaeration or post-anoxia. The operation of the AGC (enzymes and low-molecular components) in wheat (Triticum aestivum, cv. Leningradka, non-resistant to hypoxia) and rice (Oryza sativa, cv. Liman, resistant) seedlings after 24 h anoxia and 1 h or 24 h reaeration was studied. Significant accumulation of oxidized forms of ascorbate and glutathione was revealed in the non-resistant plant (wheat) after 24 h of anoxia and reaeration, indicating the development of oxidative stress. In the resistant plant (rice), reduced forms of these antioxidants prevailed both in normoxia and under stress, which may indicate their intensive reduction. In wheat, the activities of ascorbate peroxidase and dehydroascorbate reductase in shoots, and monodehydroascorbate reductase and glutathione reductase in roots decreased under anoxia and reaeration. The activity of antioxidant enzymes was maintained in rice under lack of oxygen (ascorbate peroxidase, glutathione reductase) and increased during post-anoxia (AGC reductases). Anoxia stimulated accumulation of mRNA of the organellar ascorbate peroxidase genes OsAPX3, OsAPX5 in shoots, and OsAPX3-5 and OsAPX7 in roots. At post-anoxia, the contribution of the OsAPX1 and OsAPX2 genes encoding the cytosolic forms of the enzyme increased in the whole plant, and so did that of the OsAPX8 gene for the plastid form of the enzyme. The accumulation of mRNA of the genes OsMDAR2 and OsMDAR4 encoding peroxisomal and cytosolic monodehydroascorbate reductase as well as the OsGR2 and OsGR3 for cytosolic and organellar glutathione reductase was activated during reaeration in shoots and roots. In most cases, O2 deficiency activated the genes encoding the peroxisomal, plastid, and mitochondrial forms of the enzymes, and upon reaeration, an enhanced activity of the genes encoding the cytoplasmic forms was observed. Taken together, the inactivation of AGC enzymes was revealed in wheat seedlings during anoxia and subsequent reaeration, which disrupted the effective operation of the cycle and triggered the accumulation of oxidized forms of ascorbate and glutathione. In rice, anoxia led to the maintenance of the activity of AGC enzymes, and reaeration stimulated it, including at the level of gene expression, which ensured the effective operation of AGC.

This study was conducted to identify yield and yield contributing attributes of wheat genotypes in response to heat stress. In this respect, eight wheat genotypes viz., Pavon-76, Prodip, BARI Gom-25, BARI Gom-26, BAW-1143, BAW-1146, BAW-1147 and BAW-118 were used as study materials. The experiment was evaluated in relation to heat tolerance in field condition by seeding them at November 27 (normal), December 17 (late) and January 7 (very late growing condition) over two successive years. Spike length, spike number per plant, floret number per spike and grain number per spike, floret/spike, spike dry weight and individual seed weight (seed size), grain yield, biological yield and harvest index were observed highest at normal growing condition. At late and very late growing heat stress conditions, all the genotypes showed reduced performance in those parameters at different magnitudes. Heat tolerant genotypes had higher relative values in those attributes than the MHT and heat sensitive genotype at late growing condition. Late and very late growing heat stress conditions increased the sterility in all the genotypes. HSI calculated in stress environments to assess their stress potential on the basis of grain yield. The HSI values of Prodip, BAW-1146, BAW-1147, BAW-1148 and Pavon-76 were higher than 1.0, indicating that these genotypes were susceptible to heat stress. The HSI values of the remaining three wheat genotypes indicate their various levels of tolerance to high temperature. Among them BARI Gom-25, BARI Gom-26 and BAW-1143 were tolerant to heat stress.

This study aimed to assess the nutritional status of soybean and wheat and the relationship with grain yield and biometric components after lime and phosphogypsum (GY) reapplication. Treatments consisted of two lime application methods, surface liming and incorporated liming, to different levels of base saturation (BS) (44, 70, and 90%), in addition to surface liming to 70% BS associated with a standard dose (70GY1) or double dose (70GY2) of GY. Treatments were applied in 2012 and reapplied in 2016. The results refer to the 2018/2019 soybean season and the 2019 wheat season. Lime + GY treatment increased leaf area, plant height, leaf dry matter, and stem dry matter in soybean but had no effect on specific leaf area (235.4 cm2 g−1), stem diameter (6.08 mm), number of pods per plant (71.7), grain number per plant (165), or thousand grain weight (117 g). Liming to 90% BS reduced leaf manganese contents in both crops and leaf iron content in soybean. GY application improved the calcium status of both crops and the sulfur status of soybean. However, 70GY2 reduced leaf magnesium contents in soybean and wheat. Soil tillage without lime reduced wheat yield by 12% compared with no-till. Soybean yield correlated positively with leaf nitrogen (0.79), leaf phosphorus (0.91), leaf area (0.62), and thousand grain weight (0.65), whereas wheat yield correlated with leaf phosphorus (0.83), calcium (0.75), and sulfur (0.74). For improved soybean and wheat performance, it is preferable to apply lime and GY to the soil surface (70GY1), permanently maintaining no-till practices. [ABSTRACT FROM AUTHOR]

This study evaluated the efficacy of phytogenic silver and zinc nanoparticles in improving heat resilience in various wheat varieties. The silver and zinc nanoparticles were synthesized using plant leaf extract and characterized using various techniques. Four wheat varieties (DBW187, Black Wheat, DBW 50, and PBW 621) were subjected to field trials. The random block design was used, and nanoparticles in different concentrations were applied at various growth stages and morphologically, and yield parameters were recorded. UV–vis spectroscopy spectral analysis showed peaks for Ag nanoparticles at 420 nm wavelength and Zn nanoparticles at 240 and 350 nm wavelength, depicting the preliminary confirmation of nanoparticle synthesis. Electron microscopic analysis (TEM and SEM) provided morphological insights and confirmed synthesis of fine-sized particle mostly in a range between 10 and 60 nm. Energy dispersive x-ray analysis confirmed the elemental composition of the synthesized nanoparticles, with Ag and Zn elements detected in their respective samples. It also confirmed the oxide nature of synthesized ZnNPs. Dynamic light scattering analysis provided size distribution profiles, indicating average sizes of approximately 61.8 nm for Ag nanoparticles and 46.5 nm for Zn nanoparticles. The concentrations of Ag and Zn nanoparticles in the samples were found to be 196.3 ppm and 115.14 ppm, respectively, through atomic absorption spectroscopic analysis. Fourier transform infrared spectroscopy analysis revealed characteristic functional groups present in the nanoparticles. The results of field experiments established that Ag nanoparticles at 75 ppm concentration exhibited the most significant enhancements in plant growth. Conversely, Zn nanoparticles at a 100 ppm concentration demonstrated the most substantial improvements in the growth and yield of heat-stressed wheat varieties. The study concludes that optimized concentrations of silver and zinc nanoparticles can effectively improve heat stress resilience in wheat. Fungal diseases cause significant yield loss to wheat production. Septoria tritici blotch (STB), caused by the ascomycete fungus Zymoseptoria trtici, is one of the major fungal diseases affecting wheat production worldwide. In Ethiopia, STB is a severe problem that causes significant yield loss in high and mid‐altitude wheat‐growing areas. The use of resistant varieties is one of the sustainable disease management strategies, particularly for resource‐poor farmers in developing countries. Two hundred and fifty bread wheat genotypes were evaluated to identify septoria tritici resistant genotypes and estimate the extent of genetic variability for resistance to STB and other economically important traits using alpha lattice design under natural infestation in two STB hotspot environments. Analysis of variance revealed highly significant differences among genotypes, environment, and genotype × environment interaction for all traits measured. The genetic coefficient of variance was lower than the phenotypic coefficient of variance for all traits studied, and both test environments showed the influence of the environment on trait expression. High and moderate heritability values were observed for the septoria disease severity parameters, indicating that the STB resistance trait was less influenced by the environment. The days to heading and plant height were inversely correlated with disease severity. This suggests that genotypes with tall plant height and long maturity period could be resistant to septoria tritici blotch through escape mechanisms. Four of the genotypes, namely, G‐215, G‐255, G‐257, and G‐258, were found to be resistant across all locations. These and other promising genotypes will be used in future breeding programmes to select or develop high‐yielding and STB‐resistant bread wheat genotypes that can be deployed in septoria tritici blotch‐prone areas. The resource pool diversity hypothesis (RPDH) posits that organic systems that include diverse crop rotations, green manure and organic fertilizers have a greater diversity of resource pools that can alleviate weed–crop competition compared to conventional systems. To test the RPDH, wheat (Triticum aestivum) and wild oats (Avena sterilis) in winter, and maize (Zea mays) and Palmer amaranth (Amaranthus palmeri) in summer, were grown in pots in soils from organic (ORG) and conventional (CONV) fields, with contrasting number of estimated resource pools. A replacement series design with proportions of crop: weed of 0:1, 0.25:0.75, 0.5:0.5, 0.75:0.25 and 1:0 was used. Half of the pots received an organic‐mineral fertilizer (F) and the other half did not (NF). Aerial biomass of all plants was measured. Relative Crowding Coefficient (RCC) and the product of RCC (RCCP) for each crop and weed biomass were estimated by modelling the biomass across crop: weed proportions. RCCP larger than one was used as an indicator of overyielding. In the winter experiment, wheat always outcompeted wild oats and we did not detect overyielding across fields with different number of pools, system or fertilizer. In the summer experiment, we found overyielding in ORG NF fields. Functionally different species like maize and Palmer amaranth may have been extracting nutrients from diverse pools in organic NF fields whereas with the addition of fertilizer, Palmer amaranth outcompeted maize. Biomass of monoculture crop and weed in both experiments were positively correlated with number of estimated pools, even if none of the competition indexes were. Our results provide some support for the RPHD in species that are functionally different but also question the definition and nature of the nutrient pools, as well as emphasize the need for more research in the area. Calcium-dependent protein kinase (CDPK) is member of one of the most important signalling cascades operating inside the plant system due to its peculiar role as thermo-sensor. Here, we identified 28 full length putative CDPKs from wheat designated as TaCDPK (1-28). Based on digital gene expression, we cloned full length TaCPK-1 gene of 1691 nucleotides with open reading frame (ORF) of 548 amino acids (accession number OP125853). The expression of TaCPK-1 was observed maximum (3.1-fold) in leaf of wheat cv. HD2985 (thermotolerant) under T2 (38 ± 3 °C, 2 h), as compared to control. A positive correlation was observed between the expression of TaCPK-1 and other stress-associated genes (MAPK6, CDPK4, HSFA6e, HSF3, HSP17, HSP70, SOD and CAT) involved in thermotolerance. Global protein kinase assay showed maximum activity in leaves, as compared to root, stem and spike under heat stress. Immunoblot analysis showed abundance of CDPK protein in wheat cv. HD2985 (thermotolerant) in response to T2 (38 ± 3 °C, 2 h), as compared to HD2329 (thermosusceptible). Calcium ion (Ca2+), being inducer of CDPK, showed strong Ca-signature in the leaf tissue (Ca—622 ppm) of thermotolerant wheat cv. under heat stress, whereas it was minimum (Ca—201 ppm) in spike tissue. We observed significant variations in the ionome of wheat under HS. To conclude, TaCPK-1 plays important role in triggering signaling network and in modulation of HS-tolerance in wheat. Entomopathogenic nematodes (EPNs) are well-known biocontrol agents to control both above- and below-ground insect pests. EPNs can also control plant parasitic nematodes by releasing allelochemicals through symbiotic bacteria present in their guts. This study aimed to evaluate the bio-efficacy of a novel powder formulation comprising of infective juveniles (IJs) of a heat-tolerant indigenous EPN species of India, Steinernema abbasi, against subterranean termites (Odontotermes obesus) in wheat and chickpea fields. The bioefficacy of test formulation in field study for three rabi seasons (2020-2023) in wheat and chickpea grown in field heavily infested with subterranean termites decreased the plant damage from termite attack, monitored as relative number of infested tillers in wheat and infested plants in chickpea fields. The reduced damage in test crops caused by termites increased growth, yield attributes and yield of test crops. This present research validated the prepared formulation's potential as a biopesticide ideal for organic farming and integrated pest management practices. [ABSTRACT FROM AUTHOR]

In the present study, a new approach that uses ultrasound-assisted extraction clean-up combined with dispersive liquid-liquid microextraction has been developed for the preconcentration of trace amounts of clodinafop-propargyl herbicide in wheat samples. The clodinafop-propargyl was then determined using gas chromatography with flame ionization detection. In this technique, wheat samples were extracted by ultrasound-assisted extraction followed by a dispersive liquid-liquid microextraction (DLLME) step for further purification and enrichment of clodinafop-propargyl. By using a Pasteur pipette as an extraction device, a low-density extraction solvent like water is possible to use in the DLLME method. The effects of various parameters on the extraction efficiency of the proposed technique were investigated and optimized. Good linearity was obtained from 20 to 1000 µg/kg. The limit of detection was 10 µg/kg (S/N = 3). The relative recoveries at the three spiked levels ranged from 88 to 96% with an RSD less than 13% (n = 3). Fusarium head blight (FHB) is a significantly important disease in cereals primarily caused by Fusarium species. FHB control is largely executed through chemical strategies, which are costlier to sustainable wheat production, resulting in leaning towards sustainable sources such as resistance breeding and biological control methods for FHB. The present investigation was aimed at evaluating newly identified bacterial consortium (BCM) as biocontrol agents for FHB and understanding the morpho-physiological traits associated with the disease resistance of spring wheat. Preliminary evaluation through antagonistic plate assay and in vivo assessment indicated that BCM effectively inhibited Fusarium growth in spring wheat, reducing area under disease progress curve (AUDPC) and deoxynivalenol (DON), potentially causing type II and V resistance, and improving single spike yield (SSPY). Endurance to FHB infection with the application of BCM is associated with better sustenance of spike photosynthetic performance by improving the light energy harvesting and its utilization. Correlation and path-coefficient analysis indicated that maximum quantum yield (QY_max) is directly influencing the improvement of SSPY and reduction of grain DON accumulation, which is corroborated by principal component analysis. The chlorophyll fluorescence traits identified in the present investigation might be applied as a phenotyping tool for the large-scale identification of wheat sensitivity to FHB.
